Note :  To generate GFP-expressing iNs, cells were lipofected 4 days before fixation using the Lipofectamine 3000 Transfection Kit. Briefly, 5 microliters OptiMEM was mixed with 0.75 microliters lipofectamine and then added to 25 microliters OptiMEM containing 0.1 microgram GFP pMAX plasmid (Lonza) and 1 microliter P3000 reagent. After incubation for 15 min, 50 microliters lipofection mix was added per coverslip, and the iNs were kept in culture until fixation. GFP-pmax-lipofected iNs were fixed at days 14 of differentiation. This reconstruction is from culture 14 days after GFP-Transfection. iNeurons are described as a mixture of upper layer cortical neurons (positive for CUX1; BRN2), sensory neurons (postive for PRPH, ISL1) and sensory neurons of the cortex (postive for both cortical and sensory neurons).
