Note :  CRISPR-Cas9 technology was used for engineering MECP2 KO in human ReNCells VM. Mecp2 KO was confirmed by western blotting. MeCP2 was restored in human MeCP2 KO cells. To improve circularization, a sequence of 35 nt upstream and downstream of introns was added. uc.478 were overexpressed by cloning into the BamHI site of the vector pLVX-shRNA2.
